In vitro maturation and fertilization of ova from the ovaries of cows

The possibilities of in vitro maturation and fertilization of ova collected from ovaries of slaughtered cows as well as culture of embryos for 72-78 hours are investigated in the present work. Culture of the ova was carried out in an MPM culture medium containing 20% inactivated OCS and 10 microg/ml FSH-P. After incubation for 22-24 hours the ova were fertilized with capacitated frozen semen. The embryos were co-cultured with cummulus and zona radiata cells, in granulosa cells the embryos were fixed, stained and evaluated as regards percentage of fertilization and stage of development. A total of 1820 ova were aspirated of the 465 ovaries collected. Of the latter 1085 (i.e. 4.7 ova per cow) were evaluated as suitable for maturation and fertilization. Of these 567 (52.3%) were fertilized. In 8 experiments, the fertilization rate amounted to 66.7-76.6% and in 22 to 16.6-61.8%. Of the 567 ova fertilized, 235 (41.44%) developed to the 2-cell stage, 113 (19.93%) to the 4-cell stage, 115 (20.28%) to the 8-cell stage and 104 (18.34%) to the 16-32 cell stage. Further efforts are required to reach more advanced developmental stages enabling transfer to recipient cows.
