Differential plant regeneration responses from protoplasts of mungbean [Vigna radiata (L.) Wilczek] and mothbean (V. aconitifolia Jacq Marechal)

A protocol for the isolation and culture of mungbean (Vigna radiata) root protoplasts and mothbean (V. aconitifolia) leaf mesophyll protoplasts gave plating efficiencies of 1.30 percent and 2.81 percent respectively. Protoclones of mothbean regenerated shoots/shoot meristemoids upon transfer to MS medium with 1.0 mg/L zeatin but those of mungbean failed to exhibit this response. Shoot meristemoids further developed into shoots upon transfer to MS medium with 1.0 mg/L kinetin. Excised shoots were rooted in MS basal medium lacking growth regulators. Rooted plantlets were successfully transplanted to compost and grow under glasshouse conditions. The potential for the development of a selection system for somatic hybrids of these two legume protoplast systems is discussed
