Analysis of the wild core collection with RAPD markers : case of the Colombian wild P. vulgaris

A core colletion for the wild P. vulgaris was established using both an evolutionary and an agroecological approach (Tohme et al., 1993). It consists of 100 accesions selected to cover the whole range of distribution of collected wild P. vulgaris. The core collection is being characterized using molecular markers such as RAPDs and RFLPs already mapped on the bean map in order to study the genetic structure, assess the extent of the founder effect and to determine gene flow between wild and cultivated and within and between gene pools. Of particular interest is the case of the Colombian wild P. vulgaris germplasm. Recent collections have increased the number and distribution of the Colombian populations. Two new phaseolins were identified from populations collected in the department of Cundinamarca. Furthermore, T and H phaseolin typical of the Andean germplasm were also ideantified. Preliminary RAPD analysis was performed using 91 polymorphic bands on a selected group of wild and cultivated accessions from Colombia; also included were wild accessions from the different gene pools selected on the basis of the phaseolin data. Cluster and correspondence analysis identified different groups related to the Mesoamerican, Northern Andean and Southern Andean pools as well as recombinant types. The analysis is being extended to the whole wild core collection using RAPDs and mtDNA clones to understand the relationship of the Colombian wild the either the Mesoamerican or Andean germplasm
