Fluorescence labelling of membrane-bound calcium and calmodulin during regeneration of Solanum tuberosum L. protoplasts

With chlorotetracycline (CTC) and fluphenazine (FPZ) respectively fluorescence the localization of (membrane-bound) calcium and calmodulin was observed. Freshly isolated protoplasts of dihaploid Solanum tuberosum (DH 81.71463) showed a high cytoplasm fluorescence. Later on, when cell division took place, the fluorescence was localized around the plasmatic membrane. Dividing cultures gave strong CTC and FPZ fluorescence during the first days of protoplast cultivation. Cell walls, nuclei and plastids were without fluorescence. It is concluded that no specific distribution of membrane-bound calcium and activated calmodulin or rearrangements of cellular calcium pools can be observed during cultivation of leaf mesophyll protoplasts.
