Assessment of enzyme-linked immunosorbent assay in comparison with complement fixation test for the diagnosis of subclinical paratuberculosis in cattle

The enzyme-linked immunosorbent assay (ELISA) was evaluated for its routine application as a field test to the serological diagnosis of bovine paratuberculosis. Sera absorbed with both heat killed organisms of Mycobacterium phlei and Kaolin were assayed for antibody activity to the crude protoplasmic antigen of M. paratuberculosis. The ELISA recorded sensitivity of 67.9 % for the subclinically infected cattle. Specificity was determined as 99.9 % for cattle from the noninfected herds. The values of sensitivity and specificity of the complement fixation test (CFT) were 26.3 % and 97.6 % respectively. Seroconversion of experimentally or naturally infected cattle was detectable with the ELISA several months earlier than with the CFT. The superior sensitivity and specificity of the ELISA support the hypothesis that it should replaced the CFT as the routine test of choice for the diagnosis of bovine paratuberculosis.
