Occurrence and detection of nitrate reductase in food samples

The production of nitrate reductase (NR) by microorganisms contaminating food products was monitored. The enumeration of viable microorganisms was performed in eleven food samples, the number of coliform bacteria was estimated and the ratio of bacterial strains exhibiting nitrate reductase activity was determined. Identification of selected coliform strains was performed microscopically and by biochemical tests. In all food samples the presence of N-nitrosamines was evaluated. A bacterial strain of Pseudomonas pseudoalcaligenes was chosen for the purification of nitrate reductase. Nitrate reductase isolated from this bacterium was characterized as assimilatory nitrate reductase and it was found that this enzyme is located in cytosol. Activity of nitrate reductase and the growth of bacteria was monitored.
