Excised anthers from three buds were cultured in a 60 mm Petri dish containing Cp medium supplemented with 0.01 mg/l 2,4-dichlorophenoxyacetic acid (2.4-D) and kinetin, respectively. Cultures were incubated for 8 days at 35 degrees C in the dark, then transferred to 25 degrees C with a 12-h photoperiod (cool white fluorescent light, 2000 lux). After 12 days of induction on Cp medium anthers were transferred to R1 medium supplemented with 0.1 mg/l kinetin. Well developed embryos were separated from the anthers and then transferred onto V3 hormonfree medium for development. The ploidy level of the resulted plants was determined
