Characteristics of mulberry callus proliferated by the application of brassinolide and coumarin

The effects of coumarin and brassinolide on the induction of callus in the mulberry soft-wood and its proliferation were studied. By the addition of 5 mg/l coumarin or 0.1 to 0.25 mg/l 2,4-D in the medium, calli were induced by 100% and their proliferation was marked. The application of 10n mg/l coumarin to the medium containing 2,4-D showed extensive promotive effects on the proliferation of calli tissues. On the other hand, the application of 0.1 mg/l brassinolide showed a similar effect as cytokinin. The calli tissues proliferated by brassinolide were of the compact type, initiating the tracheal tissues in the peripheral zone which was rich in starch grains. This state was considered to be the prostage of organ formation. In contrast, the calli proliferated by the application of 10 mg/l coumarin were friable and poor in starch grains in the peripheral zone
