Changes to histones in rat testes after irradiation by gamma rays

Cytological changes and concentration, total content, and composition of histones (the relative representation of the somatic histone fractions and of the specific testicular histone fraction TH1) in the cells of the seminiferous epithelium of rats on the 1st, 15th and 41st day were studied after a single whole body dose of 6 Gy by gamma-rays. Maximum changes in the testes were observed on the 41st day, on which day the weight of testes decreased by more than half. The decrease was accompanied by a drop in the number of spermatogenetic cells (spermatogonia, spermatocytes and spermatids). Regeneration of the seminiferous epithelium began at the end of the experiment, when an increased number of spermatogonia was found, due to the renewal of their mitotic activity. Changes in weight and cellular architecture were accompanied by a decrease in concentration and total content of histones. The reaction of individual histone fractions to gamma irradiation differed depending on their place and function in chromatin. The proportion of histone fractions H1, H4 and subfraction TH1 decreased and simultaneously the proportion of fractions H2A, H2B and H3 increased on the first day after irradiation. A decrease in subfraction TH1 was connected with the drop in highly sensitive spermatogonia and spermatocytes for which TH1 is specific
