In vitro propagation of Liquidambar formosana Hance

Seedlings and mature branches from Liquidambar formosana Hance were brought from Ang Khang Royal Project, Chiang Mai Province to the Faculty of Forestry for further studies. The explants needed were shoots and young branches. They were first sterilized and latter cultured the clean explants on Murashige and Skoog (MS) with growth regulators for shoot and root regeneration. The rooted plantlets were transplanted to study survival percentage. The studied was also involved the effect of subculture cycles on shoot multiplication and rooting quality. The sterilization methods, given the highest percentage of clean culture, for seedling explants was soaking in 0.2 percent mercuric chloride for 20 minutes and for explants from the mature was soaking in 10 percent clorox for 5 minutes followed by 0.1 percent mercuric chloride for 15 minutes. The explants from both the seedlings and the mature plants could be induced to produce thehighest number of shoots on the same medium, MS nutrients, supplemented with 6-benzylamino purine (BAP) and indole-3-butyric acid (IBA) at 1 and 0.1 mg/l respectively. The shoot multiplication rate of cultured seedling was increased up to the 9th subculture bycle for the 15 days culturing period and up to the 6th subculture cycle for 30 days culturing period. For seedling cultured, the shootlet obtained could be induced to form the best root quality in the MS medium of 1/6 strength witout ritamins, supplemented with IBA 5 mgil For the shoottets obtained from mature enplants could be induced to form the best quality roots in the medium of 1/6 strength of MS nutrients, without vitamins, supplemented with 2 mg/l IBA. At certain number of subculture cycles resulted in decreasing the rooting percentage of shootlets and the average number of roots per individual. The survival of the plantlets cultured from seedling explants was 100 percent when transplanted in sandy medium. The rooted plantlets could well develope and grow as normal when planted in soil.
