Genetic polymorphism in vineyard peach germplasm

Yugoslavia has a diverse population of so-called vineyard peaches, which has been little exploited for genetic improvement. The complement data on agronomic characters already gathered, a study of polymorphism of molecular markers has been undertaken. We have analysed 40 peach cultivars and 79 vineyard peach accessions for 32 enzyme systems using polyacrylamide gel electrophoresis (PAGE). Good activity and separation were achieved for all systems including ones previously reported as poorly resolved. On the basis of polymorphism revealed for twenty-one enzyme systems thirty loci were proposed. Vineyard peach accessions from Serbia (Yugoslavia) were polymorphic for all 13 loci proposed. Interestingly, 13 American cultivars appeared polymorphic for 27 loci indicating much more polymorphism than previously reported. The 19 peach cultivars and 39 vineyard peaches that revealed high heterozygote level were analysed by RFLPs (Restriction Fragment Length Polymorphism). Polymorphism was revealed with ten probes and fifteen loci were proposed. Vineyard peach accessions were polymorphic for thirteen loci. Data obtained from isoenzymes and RFLPs were used to calculate distances and similarities among the peach accessions analysed by UPGMA clustering method.
