The investigation of tissue culture methods on the conservation of terrestrial orchids

The protocorm-like bodies of Dactylorhiza baltica were used as a plant material. The protocorm-like bodies were dipped in 75 per cent alcohol and rinsed with distilled autoclaved water. This was followed by sterilization in sodium hypoclorite solution for 20 min. In total 28 meristems were exised from protocorm-like bodies and planted to modified Murashige-Skoog's medium supplemented with 0.5 mg/l NAA and 2.0 mg/l 2iP. After four weeks of culture, 60 per cent of the exiced apical meristems had started to grow while none of the explants originated from the lateral meristems expressed any tissue development. The latter ones were probably not fully differentiated and therefore incapable for morphological development
