Development of karyotyping techniques for the determination of chromosome number [of catfish]

A karyotyping technique suited to catfish was developed to determine the number of chromosomes of fish exposed to shocks for triploidy. Significantly higher percentage countable chromosomes was observed using 0.01% colchicine solution compared to that using 0.005%. However, no significant difference in percentage countable chromosomes was observed using exposure time of 6 and 8 hr in 0.01% colchicine solution. Exposure to cold temperature significantly altered the chromosome number towards triploidy from the expected 1 diploid: 0 triploid ratio. Mean percentage triploid was highest at 20 min exposure to cold temperature followed by 15 and 10 min, respectively, but no significant differences existed among the different exposure times
