Use of RAPD for classification and identification of genetic markers in Vetiveria spp. in Thailand

RAPD (Random Amplified Polymorphic DNA) technique was employed to distinguish 10 Vetiveria grass ecotypes of 2 species native to Thailand : 1. Yaa faek hom (Vetiveria zizanioides Nash.), : Songkhla, Srilanka and Suratthani 2. Yaa faek don (Vetiveria nemoralis A. Camus.) : Kampaengphet1, Loei, Nakhonsawan, Prachuapkhirikhan, Ratchaburi and Roi-et. RAPD generated, by 12 out of 114 different 10-mer oligonucleotide primers (Operon Technologies, USA), 158 discriminate banding. The average size of the amplified DNA fragments range from 0.21 to 2.0 Kb. The polymorphism of those DNA profiles, mediated by each of 12 primers of OPJ-4, OPJ-12, OPN-15, OPP-2, OPP-9, OPQ-2, OPR-2, OPR-6, OPR-12, OPR-13, OPS-11 and OPS-16 provide a sample, quick and reliable alternative to identify 10 vetiveria grass of Thailand. Among 12 primers, 4 primers including OPJ-4, OPN-15, OPR-13 and OPS-16 generated DNA product size respectively 0.6, 1.6, 0.4 and 0.55, 0.5 Kb. which enabled us to disinquish clearly between Yaa faek hom and Yaa faek don. Furthermore, 0.6 Kb. RAPD products generated by OPJ-4 primer and 0.55, 0.5 Kb. products generated by OPS-16 can use as the genetic markers for Yaa faek hom and Yaa faek don respectively, in southern blot hybridization. According to 0.6 kb. DNA probe hybridized with genomic DNA from Srilanka, Songkhla1, Songkhla2, Kampaengphet2 and Suratthani. 0.55 and 0.5 DNA probe hybridized with genomic DNA from Kampaengphet1 and Ratchaburi.
