RAPD (Random Amplified Polymorphic DNA) is useful molecular markers for rice genome analysis identifying molecular markers which are linked to aromatic gene will be a great significance in rice breeding and gene isolation. We analyzed the two DNA bulks from the aromatic and nonaromatic F2 of the cross KDML105 (aromatic)/CT9993-10-5-1M (nonaromatic) with 100 RAPD primers. One primer detected the same polymorphism between the two pool and the two parents. The result had been repeated several times and gave good reproducibility in 82 F2 individuals. The primer also amplified the same polymorphic products among the other aromatic and nonaromatic cultivars, confirming the close linkage between the marker and gene. It represents, therefore, a technically simplified approach of PCR-based selection and has direct, practical implications in rice breeding.
