A male-specific linkage map of bovine chromosome 6

Male-specific linkage maps are valuable for linkage analysis of domestic animals in which artificial insemination is wldely used. We report here a novel male-specific linkage map of bovine Chromosome 6 which consists of 12 markers markers including two loci typed by primers for ovine microsatellite loci. DNAs were obtained from 82 cattle of a single paternal half-sib family of commercial herds. Twenty-eight primer pairs for loci on bovine chromosome 6 and 10 primer pairs for loci on ovlne chromosome 6 were used for PCR amplification. Among the loci amplified by the 38 primer Pairs, 13 loci Were heterozygous in the sire. thus revealed to be informative for the linkage study. The linkage analysis in the paternal half-sib family revealed significant linkages between 12 out of the l3 loci. Minimizing crossover frequency between the loci resulted in the best gene order: OarJMP36-BM143-DIK082, TGLA37-BM4528, ILSTS097-ILST035-BMl236-BP7-BM9257-OarJMP12-BM2320. The present male-specific linkage map will be useful for mapping quantitative trait loci (QTL) as well as particular inheritable diseases in the cattle populations
