Effects of taurine on development to blastocysts of in vitro matured and in vitro fertilized bovine embryos

In the present experiments, effects of taurine on the development of bovine embryos obtained by in vitro fertilization of in vitro-matured oocytes were examined. Embryos developed to the 5-cell stage after in vitro fertilization were cultured without feeder cells in medium 199 supplemented with 1% calf serum (CS) either under 5% CO2 in air, in which 20% O2 is included, or 5% O2 5% CO2 and 90% N2. When the embryos were cultured in the medium with taurine under 20% O2 concentration, the addition of taurine into the medium significantly increased the rates of development to the blastocyst, and the rates were almost equivalen to those in embryos cultured in medium without taurine under 5% O2 concentration. Moreover, it was also found that the peak of blastocyst formation reached on Day 6 (Day 0; the day fertilized in vitro) in medium with taurine, whereas it delayed for about 24 hours in medium without taurine. However, the duration of the cell cycle was not affected by the gaseous environment of cultured embryos, especially O2 concentration. These results allow us to concluded that taurine exert beneficial effects on in vitro development of bovine embryos under conventional gaseous condition (5% CO2 in air)
