Histological effects of various dietary L-histidine levels on the midgut gland of Penaeus monodon Fabricius

Penaeus monodon postlarvae (PL50-60 days; average weight 0.52 g) were fed for 26 days with semi-purified test diets supplemented with different levels of histidine to determine the histidine requirement of P. monodon postlarvae. Prawns fed with 0.63% L-histidine semi-purified diet had the highest mean weight gain (59.9% plus or minus 4.35), whereas those fed with 0.21% L-histidine diet had the highest survival (100%). Abnormal histological changes in the hepatopancreas were observed in prawns fed with various diets except in the 0.63 g histidine-supplemented semi-purified diet, which is closest to the histidine content of prawn tissue. Ultrastructural alterations such as vesicles and eletron dense bodies became more distinct and extensive as the amount of histidine was decreased in the diet. Furthermore, prominent formations of cisternae and vesicles were observed with increased histidine supplementation
