Bacterial wilt of Patumma (Curcuma alismatifolia Gagnep.) and disease control

Fifteen strains of bacterial wilt (BW) of patumma (Curcuma alismatifolia Gagnep.) from infected rhizomes and stems collected from growing areas of Thailand demonstrated similar biochemical properties of Ralstonia solanacearum (RS) (formerly Pseudomonas solanacearum) from ginger, tomato, pepper and potato. Host range study using leaf clipping inoculation technique found typical wilt symptom on patumma, ginger, tomato, pepper and marigold but symptomless with local infection on tobacco and peanut whereas no wilt on sesame. Cross infection of patumma by RS of other hosts was successful only strains of ginger and sesame. In biovar test, all strains oxidized 3 sugar alcohols but did not utilized 3 disaccharides. Base on biochemical properties, host range and sugar utilization, all of patumma wilt strains were identified as Ralstonia solanacearum race 1, biovar 4. Control of BW of patumma in a greenhouse was carried out in infested loamy sand (LS) and sandy clay (SC) soils by amending with urea and lime (UL), bacterial antagonist (AN), UL+AN, chemical (dazomet), bleaching powder and non-treated control. In LS soil, control of BW with UL alone or UL+AN and dazomet were equally effective control (100 percent) and their rhizome fresh weights were significantly different from the other soil amendment and non-treated control. However, in SC soil, only dazomet provided the most effective control and maximum rhizome yield. Antiserum produced against membrane protein complex of RS from patumma demonstrated a high specificity and sensitive detection at 10*[2) cfu/ml by indirect immunofluorescent staining (IF). For assay of infected rhizomes, IF samples prepared by centrifugation of soaked rhizomes water and cultured in mSM-1 broth provided more accuracy and efficiency.
