Effect of aminooxyacetic acid (AOA) and glucose on vase life and postharvest physiological changes of Dendrobium JYT (Jew Yuay Tew) flowers

Effect of aminooxyacetic acid (AOA) together with glucose and sucrose on vase life and postharvest physiological changes of Dendrobium Jaw Yuay Tew (JYT) flowers were studied. The results showed that glucose gave a better result than sucrose in prolonging vase life of orchid flowers and orchid flowers held in the solution containing 4 percent glucose and 0.5 mM AOA had the longest vase life of 18.2 days with higher percentage of bud opening, more water uptake and fresh weight compared to those held in distilled water and other solutions. When pH of the solution containing glucose 4 percent and 0.5 mM AOA was adjusted from 3.2 to 4.2 and 5.2 resulted in a shorter vase life of orchid flowers. Lowering pH of glucose solution to 3.2 also increased vase life of orchid flowers but was less effective than holding peduncles in glucose 4 percent and 0.5 mM AOA solution. Dipping peduncles in Clorox solution reduced microorganism population in vase water resulting in an increased vase life of orchid flowers but was less effective than holding peduncles in the solution containing 4 percent glucose and 0.5 mM AOA. Dipping peduncles in 5-15 percent Clorox solution for 3 hrs before holding in vase solution containing 4 percent glucose and 0.5 mM AOA increased longer vase life of flowers while dipping peduncle in 20 percent Clorox solution for 3 hrs reduced vase life of flowers. Ethylene production of open florets held in solution containing 4 percent glucose and 0.5 mM AOA was lower than those held in distilled water. But their ACC content and ACC synthase activity of sepal plus petal and column plus peduncle held in the solution and distilled water were not clearly different.
