The role of oxytocin, cAMP and cGMP-dependent mechanisms in regulation of functions of porcine ovarian follicles

The aim of our work was the study of effect of oxytocin (OT) on porcine ovarian cells and of possible mechanisms of this effect. The effects of OT, inhibitors of cyclic nucleotide action (cAMP - Rp-cAMPS, KT5820, cGMP - KT5823) and of their combination with OT on IGF-I release and content of IGF-I, IGFBP-3, OT, PCNA (indicator of proliferation), ERK (MAP kinase) and CREB (transcription factor, stimulated by cAMP-dependent protein kinase A) of ovarian follicular cells cultured in vitro were examined. The stimulatory effect of OT on IGF-I production was detected. Inhibitors of protein kinase A and G activated IGF-I release. Furthermore, inhibitor of protein kinase A (KT5720) activated accumulation of CREB and suppressed accumulation of MAP kinase (ERK) in follicular cells. Inhibitors of protein kinase A and G prevented OT action on IGF-I. These observations suggest (1) involvement of OT in the regulation of IGF-I release by ovarian cells, (2) the important role of protein kinase A in control of CREB and MAP kinase and (3) that protein kinases A and G are mediators of OT action on ovarian cells
