Linamarin contents in cassava assayed by chemiluminescence

Linamarin contents from different tissues of cassava (stem, petioles, cortex, parenchyma, mature and young leaves) were determined by chemiluminescence. In this method, linamarin in each tissue extraction was treated with linamarase to liberate free glucose which was assayed using glucose oxidase/peroxidase/luminol system. After substracting free glucose present in the extract before linamarase treatment, the linamarin content was calculated. This method was compared with that of conventional spectrophotometric assay. Linamarin contents from both assays showed similar values of linamarin contents in cassava tissues used except mature leaves. Linamarin content in young leaves showed the highest level of linamarin about 10 micro mole/g fresh weight. Parenchyma contained the lowest linamarin contents (0.5 micro mole/g fresh weight). Linamarin contents in mature leaves assayed by spectrophotometry contained significantly higher levels than chemiluminescent assay. The poor recoveries was especially notable in mature leaves by chemiluninescence. This indicated that some substances was probably present in the extract from the mature leaves interfering with chemiluminescent assay.
