Response of different potato cultivars to microtuber induction

In the present studies four varieties desiree, diamant, cardinal and santa were cultured on media having different concentration of BAP (0, 0.2, 1 and 5 uM) with different sucrose levels (0, 3, 6 and 9%) with the addition of iron at 150mg/l. Complete randomized design with factorial arrangements was used to analyze the data on parameters, number of shoots per explant, number of roots per explant, shoot length (cm), root length (cm), tubering sites, number of microtuber per plant and microtuber weight (g). The objective of this study was to produce virus free microtuber by optimizing the culture media. The varieties desiree, diamant and cardinal gave the highest number of shoots per explant with 1uM 13 AP while variety Santa produced the highest number of shoots with 1uM BAP concentration. Cultivar desiree produced the maximum number of shoots per explant at 9% sucrose level but was statistically similar with varieties diamant and cardinal at 6% sucrose level. Treatment combination of BAP ( luM) with 6% sucrose level induced the maximum number of shoots per explant followed by 5uM BAP with 6% sucrose. It was observed that both varieties desiree and diamant gave higher number of roots as compared to cardinal find Santa. IuM BAP concentration generated higher number of roots per explant than other BAP concentrations. The variety desiree developed the maximum number of roots per explant at 9% sucrose level than other treatment combinations. Both varieties diamant and cardinal were, however, non- significant with desiree at 6% sucrose level. 1uM BAP concentration produced maximum number of roots per explant at 6% Soucrose level which was statistically similar to 5uM BAP concentration for number of roots per explant. The greatest shoot length was produced by diamant and cardinal with 1uM BAP concentration. Santa attained maximum shoot length at 5uM BAP. Desiree gave the highest shoot length at 9% sucrose level, which was non-significant with diamant at 6% sucrose level. Desiree, cardinal and santa produced long shoots at 6% sucrose level and were statistically at par. 1uM BAP concentration gave maximum shoot length with 6% sucrose level and with the same sucrose level. 5uM BAP concentration gave statistically similar shoot length. Variety santa generated maximum root length with 5uM BAP concentration. Diamant and cardinal were non-significant with Santa at 1uM BAP concentration. The greatest root length was produced by desiree with 9% sucrose level, which was statistically at par with santa at 6% sucrose level. 1uM BAP with 6% sucrose lever gave maximum root length which was, non significant with luM BAP with 9% Sucrose level and 5uM BAP with 6% sucrose level. All varieties produced maximum tuber on axillary positions rather terminally. Santa produced maximum tubers (96%) at axillary position followed by Desiree, diamant and cardinal 1uM BAP induced maximum number of micro tubers per plant followed by 5uM BAP concentration 6% sucrose level yielded the highest number of micro tubers per plant in all the studied varieties. The highest microtuber weight was attained with 1uM BAP concentration followed by 5uM BAP concentration that was the next best. The highest microtuber weight was yielded by 6% sucrose level in all the studied varieties.
