Study on the packed spermatozoal volume and its correlation with spermatozoal count in buffaloes

Semen ejaculated from four Murrah buffaloes were used to determine the possible application of packed spermatozoal volume in the quantity of spermatozoal density in buffaloes. Semen was collected once a week through artificial vagina for eight weeks. A portion of semen collected was used for packed spermatozoal volume determination and total sperm count. Packed spermatozoal volume was determined by filling a capillary tube with approximately 0.02 ml of fresh semen. It was centrifugated for 10 min at 10,000 rpm. Total sperm count was determined using routine hemocytometer method. Data gathered were statistically analyzed and interpreted. Correlation analysis revealed that packed spermatozoal volume and total sperm count were strongly correlated. Correlation analysis also showed that as the packed semen volume increases, there was a corresponding increase in total sperm count. It was concluded that packed spermatozoal volume, expressed in millions per millimeter, could be used in estimating the value of total sperm count. Its use in the field was recommended
