Aflatoxin analysis with minicolumn technique

The aim of this study is to adapt minicolumn technique, that is used as a qualitative method for mycotoxins analysis, to our laboratory, and also determine the detection limits of this method. For this aim 10 feedstuff and 12 mixed feed; totally 22 feed samples were used. Ten ppb, 20 ppb, 50 ppb and 100 ppb aflatoxins were added to each feed samples, respectively. After extraction of these samples, the extracts were condensed and transferred to the minicolumn. After the extract and the developing solution were passed through the column, results were evaluated under UV light to observe blue or green fluorescence. Blue fluoresrence in florisil layer evaluated as positive. So as to confirm the test, sulfuric acid (24%, in water) was added to the columns and it was observed that the blue color was turned to yellow, which characterize for aflatoxins. Results show that the 10 ppb aflatoxin can be detected in the feedstuff and mixed feed with minicolumn technique and this method can be used in routine analysis and in practical
