Alterations of rat reticulocyte (anti)oxidant status and energy metabolism influenced by hydrogen-peroxide

The aim of this study was to evaluate the role of exogenous hydrogen-peroxide (H2O2) on the (anti)oxidant status and energy metabolism of rat reticulocytes. Washed reticulocyte-rich suspensions of red blood cells (with 60-90 % of reticulocytes) were incubated under aerobic conditions (a) without additions (control) or (b) with H2O2 (1,2,5 mmol/l). Significant increase of lipid peroxidation occurs in reticulocytes incubated with 2 and 5 mmol/l H2O2. However, it is not accompanied by increased red blood cell fragility, neither by increased MetHb and Heinz bodies' formation, nor GSH level. H2O2 only in high concentration (5 mmol/l) stimulated (not significant) OPP activity. In addition, exogenous H2O2 in concentrations of 2 and 5 mmol/l, but not 1 mmol/l, induces significant changes of energy metabolism in reticulocytes of rats. It induces reduction of total and coupled oxygen consumption accompanied by stimulation of glycolysis. Due to inhibition of mitochondrial oxidative phosphorylation appears a significant decline of ATP production. However, simultaneous stimulation of glycolysis (Pasteur effect) is not suffiecient to prevent the decline of ATP concentration. Despite reduced overall ATP production and ATP concentrations, average energy turn-over was not significant changed.
