In vitro establishment of selected 'Rocha' clones (Pyrus communis L.) from nodal cuttings [Portugal]

An efficient methodology for the in vitro establishment of selected fruit tree genotypes is a valuable biotechnology tool. Experiments were carried out aiming at an improved protocol for the in vitro establishment of selected 'Rocha' clones using uninodal cuttings as explants. Cuttings were obtained from growing shoots collected early in the spring of 1998 and 1999 from the same three mature 'Rocha' trees in the field. The effects of 'Rocha' mother plant, basal salt medium (QL vs. MS) and BAP concentration (0.5, 1.0 or 2.0 mg/l) were studied. Mother plant had a significant effect on the number of shoots obtained per uninodal cutting and also on the fresh and dry weights of those shoots. The highest BAP concentration yielded the highest nimber of shoots per explant. As compared to MS, QL medium increased the fresh and dry weights of the shoots developed per explant both in 1998 and 1999 and proved to be the most adequate for the in vitro establishment of 'Rocha' from nodal cuttings
