Mass culture of Skeletonema costatum

Mass culture of Skeletonema costatum was performed under 3 experimentations. The first experiment was designed to evaluate the highest cell production by using 4 formulars of media including F1:KNO3, Na2HPO4 and Na2SiO3; F2:KNO3, NPK 16-20-0 and Na2SiO3; F3:Urea, Na2HPO4 and Na2SiO3; F4:Urea, NPK 16-20-0 and Na2SiO3. Results showed that mean of doubling time of S.costatum cultured in F1, F2, F3 and F4 were found at 0.605, 0.671, 0.735 and 0.716 day, while similar cell production were obtained in all formulars. The second experiment aims to study the 50 percent harvesting of production. Results showed the maximum cell number of S.costatum within 24 h of culture period; and doubling time in the morning harvest was better than the afternoon harvest. The third experiment was carried on to find out the optimal density of the starter cells for mass production. It was found that the maximum cell number was observed on days 2 and 3 in 1:5 and 1:10 densities of the starter cells respectively. Similarity results of the mean doubling time of cell production were obtained in both trials. It should be noted that the level of silica was dropped to zero on the second day in all tanks of this experiment.
