Development of the new methods for ecological study of soybean bacterial pustules: determination of bacteriophage as a marker for epidemiological study

Morphological feature of bacteriophage of Xanthomonas campestris pv. glycines was identical, showing polyhedral head and rod-shaped tail. Head of phage particles measured 70 * 55 mm in size and the tail 15*180 nm. The phage could be completely inactivated either by heat treatment at 60 deg C for 10 min or by diluting into pH 7.0 phosphate buffer and 90 percent ethanol. The one step growth experiment revealed that the phage had the latent period of about 50 min and average burst size of 11. The survival of phage in the environment, however, was also discussed from the viewpoint of the marker that can be used for epidemiological studies of soybean bacterial pustule in the future.
