Genetic differences among the wild isolates of Flammulina velutipes collected from a fallen tree of Broussonetia papyrifera

Genetic differences among the twelve wild isolates of Flammulina velutipes collected from a fallen tree of Broussonetia papyrifera were examined using somatic incompatibility tests, mating tests, and restriction fragment length polymorphism (RFLP) analyses of mitochondrial DNAs (mtDNAs). The results of the grouping of these isolates by their somatic incompatibility interactions, mating types and mtDNA phenotypes were all indicated the same grouping patterns, and the twelve isolates examined could be divided into four different genetic groups, namely genets, which consisted of one to five isolates. Because mtDNA from the twelve isolates showed four different RFLP phenotypes, these isolates seemed to be offsprings derived from at least four distinct genets. In addition, the isolates which belonged to the same genet were collected within the limits of about 1 m on the fallen tree. By combining the result from the present study with those of a few previous studies, it was suggested that such a distribution size of a single genet would be common occurences among wood decaying basidiomycetes including F. velutipes.
