Analysis of LGC-1, low glutelin mutant of rice

We found the rice seed protein mutants, LGC-1, which has low glutelin and high prolamin content in the EI treated progeny. Lgc-1 gene which is responsible for low glutelin and high prolamine content in LGC-1 is mapped on chromosome 2 and the distance of Lgc-1 gene from the marker XNpb243 estimated to be 8.5cM toward G365. In southern blot analysis, no difference was found in band pattern when both GluA and GluB gene were used as a probe, which indicated that there is not large deletion or duplication at glutelin genes in LGC-1. In northern analysis, it was found that GluB genes are suppressed dramatically and GluA genes are slightly suppressed in LGC-1. The result of cross between LGC-1 and transformats which has GUS gene driven by GluB promoter indicate that Lgc-1 gene dose not affect GluB gene promoter activity. To clarify the mechanism GluB genes suppression, we are now trying to isolate Lgc-1 gene.
