Clone distribution of two causal fungi of root and butt rot in a Japanese cypress (Chamaecyparis obtusa) plantation

"The distribution of clones of Phlebia chrysocrea and Tinctoporellus epimiltinus, two causal fungi of butt rot, was examined in a 25-year-old Japanese cypress plantation. The somatic incompatibility of 29 isolates of P. chrysocrea and 11 isolates of T. epimiltinus was tested on potato dextrose agar (PDA) plates and sawdust-rice bran medium. Clearer antagonism was observed on sawdust-rice bran medium than on PDA. RAPD patterns for isolates belonging to the same somatic incompatibility groups (SIGs) were identical, suggesting that these isolates belong to a single clone. All isolates of P. chrysocrea belonged to different SIGs. These findings suggest that P. chrysocrea rarely spreads by vegetative growth. Within the present sites, three isolates of T. epimiltinus belonged to the same somatic compatibility group, and 9 clones in total were found distributed. This suggests that T. epimiltinus mainly spreads by basidiospores. It was confirmed that T. epimiltinus spreads by vegetative growth, and a single clone was found to have spread to a tree 11.1 m away."
