Rapid analysis of Salmonellae contamination of chicken liver based on invA gene and 16S rDNA

Food contamination by Salmonella spp., especially Salmonella choleraesuis ssp. choleraesuis serotype Enteritidis often causes foodborne disease and human salmonellosis infection. Here, we tried to examine whether commercially available chicken liver was contaminated by Salmonella spp. based on invA gene amplification and sequence analysis of 16S rDNA. After non-selective enrichment of bacterial cells in Tripticase Soy broth, four kinds of bacterial strains were isolated by colony characteristics on deoxycholate hydrogen sulfide lactose agar plates from the sample. Two of the four strains formed black colonies induced by H2S production. It could not be confirmed whether these two strains were Salmonella spp. by their characteristics in triple sugar iron agar and lysine indole motility medium tubes. However, these strains were shown not to be Salmonella spp. by polymerase chain reaction amplification of invA gene, which almost Salmonella spp. possess. These bacteria were identified as Proteus mirabilis by sequence analyses of their 16SrDNA.
