A simple method for screening cotton germplasm against cotton leafcurl begomovirus

Cotton leaf curl disease (CLCuD) epidemic in early 90s posed aserious threat to the fibre-based economy of Pakistan. All commerciallygrown varieties showed different levels of susceptibility to the disease.Attempts to screen breeding material were hampered due to lack ofreliable screening methods. A simple lab-based technique has beendeveloped to identify resistance against Cotton Leaf Curl Virus (CLCuV)in elite commercial cotton varieties and breeder's material in the pipeline,CLCuV is transmitted by whitefly in a persistent manner therefore, thestrategy employed whitefly-mediated transmission in insect-proof cages.Susceptible genotypes produced typical CLCuV symptoms uponinoculation. Virus presence in all genotypes was tested by tripleantibody Sandwich enzyme linked immunosorbent assay (TAS-ELISA).Virus concentration in all susceptible genotypes was determined onemonth post-inoculation at ten days interval. Further indexing of symptomless and ELISA negative genotypes was confirmed by graft inoculationand Polymerase Chain Reaction (PCR). After successful graftestablishment, five genotypes (CIM 436, CIM 443, CIM 446 and CIM 448, CIM1100) produced hardly visible sporadic speck of vein thickening andwere found negative by ELISA and PCR. This technique has tremendouspotential for exploring inherent resistance in advance lines throughoutthe year.
