In vitro culture of Pachyrhizus ahipa (weed) Parodi. Effect of the application or rotenone to the medium

In preliminary in vitro culture assays of Pachyrhizus ahipa, MS medium (Murashige and Skoog) containing 1 mg/l BA, Zeatin or Kinetin single or with 0.5 mg/l IAA supplemented or not with 0.05 gr/l rotenone, was used. Rotenone is an isoflavonoide that appears in high concentration in the seeds of Pachyrhizus (Jimenez et al., 1998). For in vitro culture some pieces of actively expanding leaves, stems and leaf buds were taken from plants (AC102, AC201, AC521 and AC%"$) growing in a greenhouse. The tolerance level to rotenone was evaluated. The medium with the hormones Zeatin and BA complemented with rotenone presented better results in the growth of the stem cuts than the others. In a general way growth leaf discs of AC521 and AC102 increased when rotenone was added to the medium with Zeatin. The juvenility of the plant material is an important factor that can overcome the effect of rotenone (AC524 and AC201). The presence of rotenone in the medium does not seem to change the clorophyll content of the discs
