Molecular identification of species of the Tomato yellow leaf curl virus complex in Jordan [Lycopersicon esculentum Mill.]

anglais. The distribution of the Tomato yellow leaf curl virus complex was investigated in five locations in Jordan. Hybridization results of samples collected in 2002 showed that Tomato yellow leaf curl virus (TYLCV) occurred in all surveyed areas, while the incidence of Tomato yellow leaf curl Sardinia virus (TYLCSV) was restricted to Al-Mafraq and the central Jordan Valley. Both virus species could be detected in all areas surveyed in 2003. Using specific PCR primers, TYLCV, TYLCSV and TYLCV-Mild (TYLCV-Mld) could be detected in symptomatic tomato plants. Alignment analysis of the amplified-PCR fragments showed that the sequence of the intergenic region (IR) of TYLCSV from Jordan (TYLCSV-Jo1) shared high sequence homology with TYLCSV-ES[1] and TYLCSV-ES[2] from Spain. In addition, TYLCV-Jo1 had sequence identity of 95% and 93% with TYLCV-[Alm] and TYLCV-[DO], respectively
Afficher plus [+] Moins [-]italien. [E´ stata studiata la distribuzione del complesso del Virus dell´arricciamento fogliare giallo del pomodoro in cinque località della Giordania. I risultati dell´ibridazione dei campioni raccolti nel 2002 hanno dimostrato che il Virus dell´arricciamento fogliare giallo del pomodoro (TYLCV) era presente in tutte le aree oggetto dell´indagine, mentre l´incidenza del Virus dell´arricciamento fogliare giallo della Sardegna (TYLCSV) era ristretta ad Al-Mafraq e all´area centrale della valle del Giordano. Ambedue le specie virali sono state riscontrate in tutte le aree controllate nel 2003. Utilizzando primer PCR specifici, TYLCV, TYLCSV e TYLCV-Mild (TYLCV-Mld) sono stati identificati in piante di pomodoro sintomatiche. L´analisi di allineamento dei frammenti amplificati della PCR hanno dimostrato che la sequenza della regione intergenica (IR) di TYLCSV della Giordania (TYLCSV-Jo1) presentava un´elevata omologia di sequenza con TYLCSV-ES[1] e TYLCSV-ES[2] della Spagna. Inoltre, TYLCV-Jo1 presentava un´identità di sequenza del 93% e del 95% con TYLCV-[Alm] e TYLCV-[DO], rispettivamente.]
