Expression specificity of the globulin-1 promoter driven transgene (chitinase) in maize seed tissues [Zea mays L.]

To determine the expression specificity of transgenes driven by the seed-specific promoter of the Zea mays (maize) globulin-1 (Glb1) gene, the antifungal genes, chitinase 18 (ch18) and beta-1,3-g1ucanase II (glu) under the control of the Glb1 promoter, were bombarded into maize Type I embryogenic calli. Sixteen independent transformed lines were obtained. Transgene integration was confirmed by Southern analysis and expression detected in embryos by Northern analysis. Western analyses and enzyme activity assays showed that the ch18 transgene was expressed in embryos, in the pericarp plus aleurone and in the endosperm beginning about 14 d after pollination. Increased beta-1,3-glucanase activity was not detected. Western analysis of other plant tissues detected no CH18 protein in leaves, stems and roots of young seedlings of two different cb18 transgenic lines. However, some CH18 proteins could be detected in leaf and root tissues of older plants and in silk, cob and husk tissues. No CH18 protein was found in stems, tassels or pollen or in any tissues of non transgenic plants. These results show that the Glb1 promoter can drive transgene expression in all tissues of developing maize seeds with timing similar to that found for globulin-1 protein expression. L´analisi western di altri tessuti vegetali non ha evidenziato proteina CH18 nelle foglie, fusti e radici delle plantule di due diverse linee transgeniche cb18. Comunque, proteina CH18 è stata individuata nei tessuti delle foglie e delle radici di piante più vecchie e nei tessuti della seta, della spiga e delle brattee. Non è stata trovata proteina CH18 nei fusti, nei pennacchi o nel polline o in alcun tessuto di piante non transgeniche. Questi risultati dimostrano che il promoter Glb1 può indurre l´espressione dei transgeni in tutti i tessuti dei semi di mais in corso di sviluppo con una tempizzazione simile a quella riscontrata per l´espressione della proteina globulina-1. L´espressione può essere ritrovata talvolta anche in alcuni altri tessuti.]
