Plantlet regeneration from multiple shoots of red horse chestnut (Aesculus x carnea Hayne) by shoot tip culture and production of acclimated plants by chilling treatment

Shoot tips were harvested from a 15-year-old red horse chestnut (Aesculus x carnea HAYNE) tree and were cultured on a modified MS (Murashige and Skoog) medium with various BAP (6-benzyiaminopurine) and IBA (3-indolebutyric acid) concentrations. Shoot tips placed on the modified MS medium with 5 micro M of BAP and 0.1 micro M of IBA developed multiple shoots after 32 days of culture. The multiple shoots were transferred to a modified MS medium with 1 micro M of BAP and 10 micro M of GA3 (gibberellic acid) which produced number of shoots on the medium. The shoots were transplanted to a rooting medium which was modified WP (Woody Plant) medium with 0.1 micro M of IBA and 10g/l of activated charcoal. Fifty-three percent of the shoots rooted during the 60 days after transplanting to the rooting medium. The plants chilled for 30 days were transplanted to culture soil. They were acclimated to 17% of plants after 35 days.
