Formation of acid drainage at quarry area, 1: Isolation and identification of a becterium producing sulfuric acid

From the acid drainage of a quarry, one strain of bacteria producing sulfuric acid was isolated. This bacterium is a motile, gram-negative bacillus and forms very minute colonies less than 0.5mm in diameter on thiosulfate agar. It oxidised rapidly thiosulfate as a energy source and produced the same moles of sulfuric acid as those of the substrate. In neutral sulfur medium, after about ten-day lag phase, it began to grow and produce sulfuric acid and the pH of the medium finally dropped below 2. On the contrary, in acid medium, it did not grow and produce sulfuric acid. This bacterium could utilize neither ferrous iron as a energy source, nor nitrate as a terminal electron acceptor. Except yeast extract, tested organic compounds did not support the heterotrophic growth. But addition of them to thiosulfate medium showed various influences on the mixotrophic growth and the production of sulfuric acid. Glucose had only a slightly stimulatory effect. Glutamate, succinate and pyruvate stimulated them markedly. Citrate and acetate showed inhibitory effects. Considering these characteristics, this bacterium is tentatively identified Thiobacillus intermedius.
