Isolation of Trichoderma on selective media and its in vitro efficacyagainst Fusarium oxysporum

Trichoderma selective media (TSM), either lacking Ridomil (M1) orhaving Ridomil (M2), were used to isolate the antagonistic fungus fromwheat rhizosphere soil (S1) or fallow field soil (S2) of MalakandherResearch Farm of NWFP Agricultural University, Peshawar during 2002.Isolations were attempted using either soil plate method (P) or soil dilutionmethod (D). The antagonist was successfully isolated with both the P and Dmethods on the M1 agar medium inoculated with S1. From S2, it could onlybe isolated on M1 medium with P method. No isolation could be made fromS1 or S2 on the M2 medium by P or D methods. However, sub-culturing theantagonist from M1 onto M2 resulted in a rapid and prolific growth,showing that the presence of Ridomil inhibits Trichoderma only when itspopulation density is low. These selective media could be useful in thedetection and estimation of Trichoderma populations in soils. Ridomilconcentrations should be kept low to allow isolations from soils with lowpopulation densities of the antagonist. The isolate of the antagonist,designated as Tricho-1, suppressed the growth of Fusarium oxysporumsignificantly (P less than 0.05) on potato dextrose agar medium. It issuggested that the isolate should be tested against the potato wiltpathogen under field conditions to find out whether it can control thedisease successfully.
