Although the commercial use of controlled atmosphere (CA) storage is very extensive, the mode of action of low O2 and anoxia is not very well understood. It is possible that the mode of action of low O2 on the sensescence of horticultural crops encompasses modification of metabolic processes in addition to its inhibitory effects on ethylene action. In order to investigate this aspect of low O2 cut carnation flowers were used as a model system and the expression of alcohol dehydrogenase (ADH) and pyruvate decarboxylase (PDC) enzymes involved in the process of ethanolic fermentation under anoxia was examined.Pyruvate decarboxylase (PDC) is considered key enzyme in ethanolic fermentation and an important source of energy generation under anaerobic conditions. Alcohol dehydrogenase (ADH) is the major enzyme of fermentation in plants and is responsible for recycling NAD+during anoxia. Two partial cDNA clones (PDC1 and ADH1) had been previously isolated from carnation petals (Dianthus caryophyllus) in our laboratory. These cDNA clones were evaluated as putative molecular markers of anoxic and post anoxic responses. Both cDNAs show rapid and strong induction under anoxic conditions indicating that they can both serve as useful molecular marker of anoxic response triggering.
