In vitro propagation of sago palm (Metroxylon sagu Rottboell) through embryo rescue

anglais. Sago plant, a monocot tree, hydrophilic, hapaxanthic and producing suckers plant, grown in tropical swamplands in Southeast Asia. It is locally known as ambulong, bagsang, landing, lumbai in the Bagobo dialect. The plant accumulate a huge amount of starch in its stem, very often more than 100 kg/ tree becoming an imp;ortant source of industrial starch. Likewise, germination of sago seed is generally poor because of the presence of pericarp and sarcotesta as well as low air temperature and excessive high air temperature within the seed which reduce germination. An alternative technology through tissue culture must be developed to enable to produce an elite clonal planting ,materials en masse. Presently, in vitro propagation of sago palms is still in the experimental stage. Thus, the response of premature embryo grown in MS medium with different levels of bezylaminopurine (BAP) is the main concern of this study. The results showed that premature embryo cultured onto the MS and 5 ppm BAP medium gave better survival rate of 83.33% compared to 7 ppm, 3 ppm, and 0 ppm BAP (50%), respectively within 60 d. Likewise, MS + 5 ppm medium produced the longest(5 min) and highest number of shoots (4) per culture at 40 d.
