Nematode resistance of backcross derivatives of Oryza sativa L. crosses with Oryza glaberrima Steud. and molecular characterization of introgression

Introgression lines derived from Oryza sativa xO. Glabarrima were evaluated for tolerance to root-knot nematode Meloidogyne graminicola under controlled conditions in the phytotron in PVC tubes and in soils infested with nematode on raised seed beds. Nematode infestation affected growth of rice reducing plant height, shoot and root biomass, and leaf area index. One of the introgression lines (IR 80311-48-B-B-1) was found to be resistant to root-knot nematode. SSR analysis with 152 markers revealed exchanges of segments in all the 12 chromosomes between the parent species and 122 introgression lines from five cross combination. Hot spots with frequent exchanges of chromosome segments were identified. A molecular map consisting of 129 SSR markers was conatructed for 85 introgression lines, which could be useful in monitoring introgression from O. glaberimma into O. sativa.
