Solid state fermentation of corn cobs for laccase production by Pleurotus ostreatus

Waste corncobs were used as substrate for the production of Laccase by Pleurotus ostreatus through solid state fermentation (SSF). Laccase has great biotechnological potential for bioprocess such as biobleaching of Kraft pulp and detoxification of industrial effluents. The medium was optimized by studying the effect of varying conditions including moisture level, cultivation time, inoculum size, peptone, yeast extract (nitrogen source) and Tween-80 on SSF. Laccase production was found to be promoted by the addition of peptone, yeast extract and a surfactant, Tween-80 in SSF medium of corn cobs. The best results (13 IU/gds laccase activity) corresponded to the medium containing ground corn cobs (10 g) fermented for 72 hours at pH 7 and 25 degree C temperature with a moisture level of 70%, 20% (w/v) inoculum, 0.3% peptone. 0.4% yeast extract and 0.1% Tween-80.
