Methods for disinfection of Acidovorax avenae subsp. citrulli, causal agent of bacterial fruit blotch disease from watermelon seed

Studies on seed treatment to decrease the transmission percentage of Acidovorax avenae subsp. citrulli (Aac.) through seed were conducted. The result showed that the seed sample treated by fermented in watermelon juice for 24 hours then soaking in 1 percent HCl for 15 minutes gave the highest percentage of seed germination (92 percent). Disease evaluation at 7 days old seedling showed that the seed treatments which fermentation seeds in watermelon juice for 24 hours and followed by soaking in 1 percent HCl for 15 minutes, soaking seed in 5 percent acetic acid, soaking in sterile water for 1 hour and followed by heating in microwave oven at 60 deg C - 80 deg C for 5 minutes, or treating seed at 80 deg C, proved to be more effective in reducing seed transmission of Aac. than other treatments. They gave the lowest number of infected seedling (0.82 percent). At 14 day old seedling, the treatment of fermented seed in watermelon juice for 24 hours and followed by soaking in 1 percent HCl for 15 minutes proved to be the most effective treatment in reducing seed transmission of Aac., however, the percentage of infected seedlings of 2.56 percent was still recorded.
