[Different enzyme systems capability for potato variety identification in vitro]

Research results of determination of possibilities of izozyme of acid and alkaline phosphatase, peroxidase and esterase application for the variety identification of equal-aged potato (Solanum tuberosum L.) plants in vitro were presented. Investigation of polymorphism possibilities of studied enzyme zymogrames for the variety identification as well as determination of potato organs (roots, leafy tops) suitability for the variety identification was realized. In course of study isozymes of four enzymes (esterase, peroxidase, acid and basic phosphate) were used for variety identification of 30 Belarusian cultivars. Soluble proteins were isolated from leafy tops and roots in vitro. Zones of electrophoretic activity were revealed in esterase, peroxidase and acid phosphate. Six useful loci proved to be suitable for potato variety identification: one - acid phosphate and five - peroxidase. Polymorphism and variety specificity were typical for the electroforetic spectrum of peroxidases of similar age plants in vitro cultivated in the similar conditions. Application of izoperoxidase spectra could solve the problem of potato variety identification in vitro because no variety showed the izozyme spectrum identical to the other ones. The highest specificity was noted in the peroxidases of potato leafy tops. The revealed locus of isozyme acid phosphase did not have enough polymorphism for variety identification but could be used in complex with other isoenzyme systems
