[Isolation and characterization of chitinase produced by Trichoderma viride TNJ 63]

Trichoderma viride TNJ63, a fungus isolated from citrus plantation soil in Riau, is able to produce chitinase activity in a significantly higher amount (p less than 0.05) than that produce by other strains of Trichoderma isolated from various citrus, cocoa and pepper plantations in Riau and Lampung. Using specific substrates, three types of chitinase activity was detected. An endochitinase produced by Trichoderma viride TNJ 63 could be separated from N-acety-betha-D-glucosaminidase and 1,4-betha-chitobiosidase activities by dialysis of the crude filtrate, followed by gel filtration chromatography after concentration by polyethylene glycol 35.000. The endochitinase has an optimum pH of 5.5 and optimum temperature at room temperature (30 deg. C)
