Susceptibility of different age of tiger grouper, E. fuscoguttatus to the iridovirus infection

The aim of the experiment is to know susceptibility of different age of tiger grouper, E. fuscoguttatus to the iridovirus infection. Fish used were larvae of 20 and 28 days old and juveniles of 3 and 5 months old. Twelve fish for larvae and juvenile of 3 months old were immersed in different concentration of iridovirus inocula for 6 hours: 1 ml/liter sea water (A), 0.1 ml/liter sea water (B) 0.01 ml/liter sea water (C), and control (pbs) (D). Meanwhile, ten fish for juvenile of 5 months old were injected intramuscularly with different dosage of iridovirus inocula, i.e: 10 ml/kg bw (A) 1 ml/kg bw (B) 0.1 ml/kg bw (C), and control (D). The experiments were conducted separately for each age in completely randomized design with three replications. The cause of mortality was also checked by PCR and histopathology. The results showed that clinical signs of diseased fish including lost of appetite, weakened, resting on the bottom were observed. Average survival for larvae of 20 days old were 13.35 percent for (A), 21.65 percent for (B), 43,35 percent for (C) and 85.00 percent for (D), larvae of 28 days old were 26.67 percent for (A) 45.00 percent for (B) 56.67 percent for (C) and 88.40 percent for (D), juvenile of 3 months old were 57.00 percent for (A), 81.67 percent for (B), 93.40 percent for (C) 100.0 percent for (D), and juvenile of 5 months old were 16.67 percent for (A), 46.67 percent for (B), 60.00 percent for (C), and 100.0 percent for (D). The dead fish from treated groups were gave positive (virus infection) results with PCR test. Histopathological observation revealed the enlarged cells in spleen and kidney tissue.
