Identification of species specific marker(s) in ruminants using mini-/micro-satellite associated sequence amplification (MASA).

Oligonucleotides primers(3) commonly used for classical DNA fingerprinting were utilized as single primer to generate mini-/micro-satellite associatedsequence amplification(MASA) profiles among the four rminants (cattle, buffalo, sheep and goat) using step down (SD)-PCR. The primers (033.15,033.6 and OAT 18.2) generated total of 41 amplicons on PAGE, outof which 36 were polymorphic.The average number of bands ranged from 4.00 to 13.00 on PAGE with size range of 285 to 2290 bp. Microsatellite primer (OAT 18.2) generated higher number of bands than the minisatellite (O33.15,O33.6) primers. All the three primers produced several species-specific bands. Between species similarity estimates from the O33.6 primer was observed to be maximum for sheep vs. goat (0.696) and cattle vs. buffalo (0.667). Genetic reltedness estimates from the microsatellite (OAT 18.2) primer revealed a close relationship of buffalo with sheep and goat rather than the cattle. In the present study, the application of SD-PCR for MASA in ruminants is being reported for the first time.
