Effect of growth regulators, Agar concentration and scale segment on Lilium ledebourii micropropagation in spring season

Lilium ledebourii is one of the beautiful species of lilium genus. This specie has high potential foe export. In this study the effect of different factors on Micropropagation were considered. The plant bulbs were collected in Damash region (Gilan province). Middle scales (1x1 cm) were used as explant. The scales were sterilized in 70% ethanol for 5 second and 2% (w/v) sodium hypochlorite solution for 30 min and were cultured on a MS medium consisting 4 levels hormone treatments : T1: no hormone, T2: 0.5 mg/lit BA + 0.01 mg/lit NAA, T3: 0.1 mg/lit BA + 0.1 mg/lit NAA, T4: 0.5 mg/lit BA + 0.5 mg/lit NAA and 3 levels of sucrose (30, 60 and 90 g/lit) with 7 g/lit agar. Scales divided to two segment (distal and basal) before culture. This research were studied using a factorial experiment based on Randomized complete design with 3 replications and four sample for every treatment. Analysis variance showed effect of sucrose concentration and scale segment were significant for bulblet number in each explant. The most bulblet number was related to 30 g/lit sucrose and it decrease, with increasing of sucrose concentration. Effect of hormone and scale segment were significant for bulblet weight and the most weight was related to T3. Effect of scale segment were significant for bulblet height, root number and root length and the basal scale showed the best response. Bulblets were transferred to bed component ¼ Tea waste compost + ¼ Peat + ¼ Leaf mold + ¼ Sand in green house after cold treatment.
